Effect of Dietary Restriction on Hepatic Antioxidant Enzyme and Reactive Oxygen Species-Generating Enzyme Activities in the Aging Process of B/W Mice

Abstract
The present study was undertaken to investigate the anti- aging effect of dietary restriction(DR) in the liver of B/W mice. Reactive oxygen species(ROS) generations in mitochondrial and postmitochondrial fraction increased with age, while they significantly decreased by DR. Glutathione (GSH) levels in mitochondrial and postmitochondrial fractions decreased as they age, whereas they were significantly increased by DR. The activities of antioxidant enzymes such as superoxide dismutase, catalase, GSH peroxidase, GSH reductase and GSH S-transferase were increased by DR. Xanthine oxidase(XOD) and cyclooxygenase(COX)-derived free radicals increased with age, while they were reduced by DR. There results suggest that DR inhibits ROS generation from XOD and COX, and maintains activities of antioxidant enzymes.
